Unless the girl is really, really quiet, then if you can't tell she is interested then typically they aren't. I call usually tell right away whether I have a shot with the girl, just by look them in the eyes. If they turn away all shy like or start staring at you back (smiling usually) they are potentially interested. But if they seem to ignore eye contact or look at you weirded out then they aren't interested. People are easy as hell to read once you know what to look for.
