how you do your portfolio depends very much on what you are doing it for.

e.g. most contests want slides, most face to face meetings with an art director will require large prints (but you might also need slides), getting a fine-art exhibition will probably require a set of fine prints ready for framing, and for soliciting people you meet to model for you, all you really need are some nice postcards or a flipboox of 4x6 prints.

I highly recommend the book The Photographer's Market Guide to Photo Submission and Portfolio Formats by Michael Willins. It has a wealth of information on how to present your photos in a variety of different situations. I don't know of any websites that are anywhere near as thorough as that book.
