Holy shenanigans!
I just had the longest day ever but i think it went well. I will find out tomorrow! today was incredibly productive, due to the fact that all of my experiments from last week didnt work. As a troubleshooting test, i went through then entire set of experiments, save for the spleen extraction and b cell/tcell purification.
i did:
PCR on old cDNA
RNA extraction (so great, got 2 micrograms/microliter)
dnase treatment
Reverse transcriptase (new cDNA)
did another PCR on new cDNA
did a Real Time Quantitative PCR on new cDNA
although it was really productive, i know see how i was able to do this.
its effin 250 in the morning and i just got home
luckily, i can sleep in tomorow.

it didnt fuckin work. no, no pcr product. no qpcr product! where the hell is my cdna???!!?

honey, this blog is kinda freakin' me out. it is pumpkin and apple pie...but everyone was so full they barely ate it, ah well, breakfast tomorrow.
